| 體外研究 | 天蠶素A顯示出抗癌活性。 Cecropin A(10-50μM)劑量依賴性地降低HL-60細胞的活力。 Cecropin A(30μM)促進ROS產生,導致線粒體膜電位(Δψm)崩潰,并在HL-60細胞中產生核染色質的形態變化。 Cecropin A(30μM)也可導致早期凋亡,并導致HL-60細胞中不依賴caspase的細胞死亡[1]。天蠶素A對革蘭氏陰性細菌具有細胞毒性,包括鮑氏不動桿菌(CCARM 12005,CCARM 12035,CCARM 12036,CCARM 12037),其最小抑制濃度(MIC)為0.5-1μM。 Cecropin A(25μM)顯著阻斷mTNF-α,mIL-1β和mMIP-2 mRNA的表達,并略微抑制RAW264.7細胞中mMIP-1 mRNA的表達。 Cecropin A(0.1,0.25,0.5,1,2.5,5μM)也抑制NO的產生并降低LPS刺激的RAW264.7細胞中的mTNF-α細胞因子水平,并且具有抗炎活性[2]。 |
| 細胞實驗 | 簡而言之，將補充有10％熱滅活FCS的RPMI 1640中的5×10 5個細胞/ mL置于96孔板上。將天蠶素A加入細胞培養物中，終濃度為10,20,30,40和50μM，將細胞在37℃，5％CO 2的潮濕氣氛中培養24小時。然后，向每個孔中加入20μLMTT（0.5mg / mL），并將板在37℃下溫育4小時。除去MTT溶液，向每個孔中加入含有0.04N鹽酸的異丙醇以溶解甲crystals晶體。在分光光度微板讀數器上在550nm的測試波長和620nm的參考波長下測定吸光度。在不存在天蠶素A（未處理的細胞）的情況下孵育的細胞的吸光度設定為100％。結果表示為細胞活力的百分比[1]。 |
